IGF-1 LR3 (Long R3 Insulin-Like Growth Factor-1) is a modified analogue of native IGF-1, engineered with an arginine substitution at position 3 and a 13-amino acid N-terminal extension that significantly reduces binding to IGF binding proteins. This structural modification extends its half-life to approximately 20–30 hours and increases its bioavailability, making it one of the most potent and widely used growth factor peptides in cellular proliferation research.

Mechanism
IGF-1 LR3 binds to the IGF-1 receptor, activating the PI3K/Akt and MAPK/ERK signaling cascades that drive cellular proliferation, protein synthesis, and inhibition of apoptosis. Its reduced affinity for IGFBPs means a greater proportion remains in its active, unbound form — resulting in stronger and more sustained receptor activation compared to native IGF-1.
